Vitamin B6: what it does and why it's key for the brain

Vitamin B6 (pyridoxine) is an essential water-soluble vitamin that the body cannot produce on its own. It participates in more than 100 enzymatic reactions, but its most relevant role for mental wellbeing lies in the synthesis of neurotransmitters.

What vitamin B6 does

B6 is an essential cofactor in the production of:

  • Serotonin — the mood neurotransmitter, made from tryptophan
  • Dopamine — motivation and reward
  • GABA — calm and relaxation
  • Noradrenaline — stress response and attention

Without enough B6, the body has the raw material (amino acids) but cannot convert it into the messengers it needs. It's the difference between having the ingredients and having the recipe.

Vitamin B6 and premenstrual syndrome

One of the best-evidenced uses is the reduction of premenstrual symptoms, especially irritability, mood changes and fatigue. Studies show significant improvement at doses of 50–100 mg/day, possibly due to its role in serotonin and dopamine synthesis.

B6 and cognitive function

Together with B12 and folate, B6 helps control homocysteine levels — an amino acid that at elevated concentrations is associated with cognitive decline and increased cardiovascular risk. This trio of B vitamins is one of the most researched supplements for brain health in older adults.

Signs of B6 deficiency

Severe deficiency is uncommon, but subclinical deficiency (low levels without clear symptoms) is more common than it might seem, especially in people with high energy demands, chronic stress or a poorly varied diet. Signs include: irritability, fatigue, difficulty concentrating, and in more advanced cases, dermatitis and anaemia.

How to take vitamin B6

The recommended daily intake is 1.3–1.7 mg for adults. For supplementation targeting specific effects (PMS, cognitive function), doses between 10 and 100 mg are used. The P-5-P form (pyridoxal-5-phosphate) is the active form and has better bioavailability than standard pyridoxine.
